It is intended to provide a novel endo-beta-(1->6)-galactanase, its gene, etc. which are expected as useful in clarifying the functions of AGP having various physiological functions such as regulation of pollen tube growth and regulation of reproduction of plants and, therefore, applicable to the construction of transformed plants (male sterility, etc.), food processing, drugs and enzymes for constructing protoplasts. beta-(1->6)-galactanse is purified 57-fold from Onozuka R-10, which is a marketed cellulase preparation, and analyzed by using galactan originating in Prototheca zopfii. As a result, it shows the peak activity at pH 4.3 and specifically hydrolyzes beta-(1->6)-galactooligosaccharides with degree of polymerization of 3 or higher and derivatives having a 4-O-methyl-glucosyluron or a glucosyluron group at the nonreducing end. Next, the full-length cDNA of this enzyme is prepared by the 3'- and 5'-RACE-PCR with the use of T.viride spore-origin RNA as a template.
