Disclosed are analogues of cyclosporin-A comprising modifications of the substituents at the positions of amino acids 1 and 3, according to Formula L. The disclosed compounds have affinity for cyclophilin, and reduced immunosuppressivity in comparison with cyclosporin-A and analogs thereof modified solely at position 1, thereby being useful in the treatment of cyclophilin mediated disease and injury including viral infection, cancer and inflammatory disorders. Also disclosed are methods of synthesis of the disclosed compounds.
