The present invention is directed to a method of activating expression of a target gene in a cell involving the treatment with at least one transcription activator like effector (TALE) polypeptide and with at least one epigenetic inhibitor substance, and an isolated cell obtainable by said method. The present invention is further directed to the use of said substances for activating expression of a target gene in a cell as well as to a composition or kit-of-parts comprising the same.
