The present invention is directed to a mediator-antibody covalent attachment method that can be activated only after binding with a binding site on the mediator-avaiable antibody. To this end, there is provided herein a mediator that is engineered to contain a photoreactive functional group at one or more specific positions (s) on the mediator so as not to interfere with the specific non-covalent molecular interactions with the binding site on the mediator-But still allow and improve covalent bond formation.
