PURPOSE: A sustained-release pharmaceutical composition including mosapride or the salt thereof are provided to rapidly reach to the effective blood concentration of the mosapride at the beginning and sustain the effective blood concentration of the mosapride at the end, thereby enhancing drug effect of the mosapride by administering once a day.CONSTITUTION: A sustained-release pharmaceutical composition comprises mosapride or the salt thereof and a pharmaceutically allowable carrier. When the composition is measured at 37 deg. Celsius according to the second rule of elution in the Korea pharmacopoeia, the following elution profile is satisfied: releasing 10-50% of the total weight of the mosapride or the salt within half an hour and accomplishing the emission of 50-95% of the total weight of the mosapride or the salt between 6-16 hours. A controlled release comprises 20-40% of the mosapride or the salt based on the total weight and the sustained-release contains 60-80% of the mosapride or the salt thereof based on the total weight. 5-80 weight% of the release controller is included based on the total weight of the sustained-release. The composition is manufactured into powders, pellets, capsules, tablets, coated tablets, or a combination thereof. The tablet is bilyer tablet which is composed of controlled release layer and sustained-release layer. The tablet includes sustained release coating pellets included within matrix.
