Disclosed is a method for cloning a λ (lambda) light chain variable region nucleic acid sequence, comprising the steps of: (a) obtaining a cell from a mouse comprising an endogenous λ light chain allele comprising an unrearranged human Vλ gene segment and an unrearranged human Jλ gene segment, where the mouse comprises a deletion of a first Vλ-Jλ-Cλ gene cluster of the endogenous mouse λ light chain allele and replacement, in whole or in part, of the Vλ-Jλ gene segments of a second gene cluster of the allele with the human Vλ and Jλ gene segments which are operably linked to a mouse λ constant region (Cλ) gene so that the mouse expresses a light chain comprising a human λ variable domain and a mouse Cλ domain; and (b) cloning from the cell a λ light chain variable region nucleic acid sequence derived from the endogenous λ light chain allele.
