The present invention disclosed an anti-cancer compound comprising a cleavable linking group that is specifically activated by the tumor microenvironment and uses thereof. The anti-cancer compound formula is as shown below, where R1 is a general functional group or protecting group； R2 is Ala, Thr, Val or Ile； R3 is Ala, Thr , Val or Asn； and R4 is a hydroxy or amino linked drug group, and the general formula for the drug is R4-H. The compounds are only activated locally in the tumor, not only avoiding the disadvantage that conventional chemotherapeutic drugs damage the immune system, but also promote anti-tumor immunity by removing tumor immunosuppressive cells.
