PROBLEM TO BE SOLVED: To provide: methods of inhibiting wild-type and certain mutant forms of human histone methyltransferase EZH2； and methods of treating cancers including follicular lymphoma and diffuse large B-cell lymphoma, or methods for identifying small molecule selective inhibitors of the mutant forms of EZH2 and methods for determining responsiveness to an EZH2 inhibitor in a subject.SOLUTION: A method for treating or alleviating a symptom of a cancer or precancerous condition in a subject comprises administering a therapeutically effective amount of an EZH2 inhibitor to a subject expressing a mutant EZH2 comprising a mutation in the substrate pocket domain in a specific sequence.
