A method of treating an inflammatory disease is disclosed. The method comprises administering to the subject a therapeutically effective amount of a polypeptide comprising a pro-domain of TNF-alpha converting enzyme (TACE), said polypeptide being devoid of a catalytic domain of said TACE, said polypeptide comprising a modification at a site selected from the group consisting of R58, R56 and K57 which renders said polypeptide resistant to furin degradation said polypeptide being capable of downregulating an activity of TACE, thereby treating the inflammatory disease.L'invention concerne une méthode de traitement d'une maladie inflammatoire. La méthode comprend l'administration au sujet d'une quantité thérapeutiquement efficace d'un polypeptide comprenant un pro-domaine de l'enzyme de conversion de TNF-α (TACE), ledit polypeptide étant dépourvu d'un domaine catalytique dudit TACE, ledit polypeptide comprenant une modification au niveau d'un site choisi dans le groupe consistant en R58, R56 et K57 qui rend ledit polypeptide résistant à la dégradation de furine, ledit polypeptide étant apte à réguler à la baisse une activité de TACE, traitant ainsi la maladie inflammatoire.
