The invention additionally provides methods for preparing and using such macrocycles, for example in therapeutic applications. In one aspect, the invention provides a method for preparing a purified peptidomimetic macrocycle composition. In one embodiment, the method comprises contacting a peptidomimetic precursor and purifying crude peptidomimetic macrocycles to obtain the purified peptidomimetic macrocycle compositions.
