The present invention relates to a sustained-release preparation which limits release and absorption of drug using a gastroretentive system and which can minimize the effect of in vivo environment, and to a method for preparing the sustained-release preparation. More particularly, the sustained-release preparation according to the present invention comprises a swellable polymer, and a first controlled-release base and a second controlled-release base. Therefore, the sustained-release preparation of the present invention may adjust the degree of swelling and floating of a polymer in order to effectively achieve gastric retention. Thus, drugs may ultimately be retained in the gastrointestinal tract over a long period of time, and the bioavailability of drugs may be improved in order to enable once-a-day administration.Linvention concerne une préparation à libération contrôlée qui limite la libération et labsorption dun médicament au moyen dun système gastrorétentif et qui peut minimiser leffet dun environnement in vivo, et le procédé de préparation de la préparation à libération contrôlée. Ladite préparation à libération contrôlée de linvention comprend en particulier un polymère pouvant gonfler, une première base de libération contrôlée et une seconde base de libération contrôlée. En conséquence, la préparation à libération contrôlée de linvention peut ajuster le degré de gonflement et de flottement dun polymère afin deffectuer efficacement une rétention gastrique. Les médicaments peuvent donc finalement être retenus dans le tractus gastro-intestinal pendant une longue période, et la biodisponibilité des médicaments peut être améliorée afin de permettre une prise quotidienne unique.본 발명은 위체류 시스템을 이용하여 약물의 방출 및 흡수를 제한하는 체내 환경의 영향을 최소화할 수 있는 서방성 제제 및 이의 제조방법에 관한 것이다. 보다 상세하게, 본 발명에 따른 서방성 제제는 팽윤성 폴리머 외에 제1 제어방출기제 및 제2 제어방출기제를 포함함으로써, 위체류를 효과적으로 달성하기 위한 폴리머의 팽윤 및 부유 정도를 조절하여, 이를 통하여 궁극적으로 약물이 위장관 내에서 장시간 체류할 수 있게 하고, 약물의 생체 이용률을 향상시켜 1일 1회 투약이 가능하도록 하는 효과가 있다.
