The present disclosure provides for an expression vector comprising (i) at least one nucleic acid sequence encoding a transcriptional gene silencing element and (ii) at least two other nucleic acid sequences selected from the group consisting of a nucleic acid sequence that encodes a nucleic acid molecule that inhibits an HIV co-receptor a nucleic acid sequence that encodes an HIV fusion inhibitor protein a nucleic acid sequence encoding an inhibitor of HIV replication and a nucleic acid sequence encoding an inhibitor of viral entry. The combination of the three mechanisms in a single expression vector provides for novel means of inhibiting HIV-I.
