the present invention is a method of preparing a matrix (stationary phase) of guar gum for use in affinity chromatography using glutaraldehyde as a crosslinking agent to produce a solid material that can be processed or fragmented with application in the field of protein biochemistry in order to obtain a material that loses its hydrogel characteristics (without water), which allows better handling for packaging in a chromatographic column, in addition to improving the flow control and the quality of the insulation process of lectins by chromatography in the washing and elution steps.a presente invenção trata-se de um método de preparo de uma matriz (fase estacionária) de goma guar para uso em cromatografia de afinidade utilizando o glutaraldeído como agente de reticulação (ou crosslinking) para produzir um material sólido que pode ser processado ou fragmentado com aplicação na área de bioquímica de proteínas visando a obtenção de um material que perde as características de hidrogel (sem água), o que permite uma melhor manipulação para empacotamento em coluna cromatográfica, além de melhorar o controle do fluxo e a qualidade do processo de isolamento de lectinas por cromatografia nas etapas de lavagem e eluição.
