The present invention relates generally to pharmaceutical therapies for the characterization of human genetic diseases, in particular unbalanced nucleotide pools, more specifically mitochondrial DNA depletion syndrome, and more particularly, thymidine kinase 2 (TK2) deficiency . The pharmaceutical regimen entails administration of at least one deoxynucleoside, or a mixture thereof. For the treatment of TK2 deficiency, the pharmacological therapy involves the administration of either deoxy thymidine (dT) or deoxycytidine (dC), or a mixture thereof.
