PURPOSE: A method for subculturing Gastrodia elata protocorms is provided to overcome Gastrodia elata protocorms degeneration. CONSTITUTION: A method for subculturing Gastrodia elata protocorms comprises: a step of germinating protocorms from seeds of Gastrodia elata using Mycena spp.； and a step of growing or enlarging the germinated protocorms using Armillaria mellea. A medium for subculturing contains Hyponex powder(2g/L) as an inorganic nitrogen source, peptone(1g/L) as an organic nitrogen source, and sucrose(20g/L) as a sugar source. The length of the protocorm is 0.8 cm.본 발명은 애주름버섯(Mycena spp.)을 발아균주로 사용하여 천마종자로부터 원구경을 발아시키는 단계； 및 뽕나무버섯(Armillariamellea)을 생장균주로 사용하여 상기 발아된 원구경을 비대 또는 생장키는 단계； 를 포함하는 천마종자로부터 종마 또는 자마를 기내배양시키는 것을 목적으로 하는 천마 원구경 계대배양방법에 관한 것이다.바람직하게는 상기 계대배양배지의 조성은 무기질소원으로 Hyponex powder(2g/l), 유기질소원으로 Peptone(1g/l) 및 당원으로서 Sucrose(20g/l)를 포함하는 것을 특징으로 할 수 있고, 상기 발아된 원구경의 길이는 0.8cm인 것을 특징으로 할 수 있으며, 상기 계대배양의 배양기간은 21일인 것을 특징으로 할 수 있다.
