The present invention provides an isolated fully human monoclonal anti-HIV antibody, wherein said antibody is PGC14 having a heavy chain comprising the amino acid sequence of SEQ ID NO: 37 and a light chain comprising the amino acid sequence of SEQ ID NO: 38. The heavy chain CDRs of the PGC14 antibody have the following sequences per Kabat and Chothia definitions: SGYSFIDYYLH (SEQ ID NO: 116), LIDPENGEARYAEKFQ (SEQ ID NO: 117), and AVGADSGSWFDP (SEQ ID NO: 118). The light chain CDRs of the PGC14 antibody have the following sequences per Kabat and Chothia definitions: SGSKLGDKYVS (SEQ ID NO: 120), ENDRRPSG (SEQ ID NO: 121), and QAWETTTTTFVF (SEQ ID NO: 44). Furthermore, the invention provides a method for producing this broadly neutralizing antibody (bNab), comprising (i) culturing an immortalized B cell clone expressing the antibody and (ii) isolating the antibody. Moreover, the invention provides a pharmaceutical composition comprising the antibody of the invention and at least a pharmaceutically acceptable carrier. According to the invention, the antibody can be used in, e.g., immunizing or reducing the effect of an HIV-infection or an HIV-related disease or for post-exposure prophylaxis； in a method of screening polypeptide libraries for polypeptides that can induce an immune response against HIV； or in immunizing an individual against a plurality of HIV-1 species.
