The present invention relates to single domain antibodies comprising at Ieast one modification relative to the 4D5 antibody scaffold or human germline VH3 domain, the modifications selected from the group consisting of H35D, A78V, S93V, S93G and W103R, with the position numbering being according to the Kabat numbering scheme. Disulfide-free variants further comprise at Ieast one additional modification selected from the group consisting of C22S, A24I, A24L and C92T, and with the proviso that at Ieast one of C22S and C92T is present. Further encompassed are the multi-modular antibody molecules and antibody conjugates comprising single domain antibodies, as well as methods for producing them. The invention in particular provides a Iibrary of the single domain antibodies or multi-modular antibody molecules and a method for selecting an antibody that binds an antigen.La présente invention concerne des anticorps à domaine unique comprenant au moins une modification par rapport au domaine VH3 de léchafaudage de lanticorps 4D5 ou de la lignée germinale humaine, les modifications étant choisies dans le groupe constitué par H35D, A78V, S93V, S93G et W103R, la numérotation des positions étant conforme au schéma de numérotation de Kabat. Des variants exempts de disulfure comprennent en outre au moins une modification supplémentaire, choisie dans le groupe constitué par C22S, A24I, A24L et C92T, et à condition quau moins une modification parmi C22S et C92T soit présente. Linvention concerne en outre des molécules danticorps multimodulaire et des conjugués danticorps comprenant des anticorps à domaine unique, ainsi que des procédés pour leur production. Linvention concerne en particulier une bibliothèque des anticorps à domaine unique ou des molécules danticorps multimodulaire et un procédé pour choisir un anticorps qui se lie à un antigène.
