The present invention relates to a method for screening an anti-cancer drug to inhibit combination between AIMP2-DX2 and HSP70 and, more specifically, to a method for screening an anti-cancer drug and an anti-cancer composition including the anti-cancer drug, which is selected by the method for screening the anti-cancer drug, as a valid component. The method for screening the anti-cancer drug comprises: a step of touching the AIMP2-DX2 or a fragment of the AIMP2-DX2, and the HSP70 or a fragment of the HSP70 with a test material or without the test material a step of measuring the combination of the AIMP2-DX2 and the HSP70 a step of determining a change of a combination level of the AIMP2-DX2 and the HSP70 by using the test material a step of selecting the test material to reduce the combination level of the AIMP2-DX2 and the HSP70 and a step of confirming anti-cancer activity of the selected test material in a cell or an animal. The present invention is useful to develop an anti-cancer drug of a new action mechanism based on a discovery that the HSP70 is a controller for stabilization by directly combining the AIMP2-DX2 which is important to cancer generation and cancer progress.
