The present invention relates to a calpain inhibitor for use for delaying or inhibiting the secondary resistance to an anti-cancer agent in a patient suffering from colorectal cancer. The invention also refers to a method for monitoring the response to a treatment of a patient suffering from a cancer chosen from colorectal cancer, gastric cancer, esophageal cancer, non- small-cell and small-cell lung cancers, leukemia, lymphomas, central nervous system maligant gliomas and ovarian carcinomas, and preferably colorectal cancer, comprising: a. treating said patient with an anti-cancer agent for a time period of at least 5 months then b. measuring the level of expression of at least one gene selected from the group consisting of calpain, annexin A2 and S100A10 genes, in cancer cells of said patient.La présente invention concerne un inhibiteur de calpaïne pour utilisation pour retarder ou inhiber la résistance secondaire à un agent anticancéreux chez un patient souffrant de cancer colorectal. Linvention concerne en outre un procédé pour surveiller la réponse à un traitement dun patient souffrant dun cancer choisi parmi le cancer colorectal, le cancer gastrique, le cancer de lœsophage, le cancer du poumon à grandes cellules et à petites cellules, la leucémie, des lymphomes, des gliomes malins du système nerveux central et des carcinomes ovariens, et de préférence le cancer colorectal, comprenant : a. le traitement dudit patient avec un agent anticancéreux pendant une durée dau moins 5 mois puis b. la mesure du niveau dexpression dau moins un gène choisi dans le groupe constitué des gènes de la calpaïne, lannexine A2 et S100A10, dans des cellules cancéreuses dudit patient.
