PROBLEM TO BE SOLVED: To provide a method for producing a contrast agent for optical imaging in which the residual amount of a single organic coloring material not bonded to albumin is small, whereas the ratio of a conjugate of albumin and an organic coloring material is large.SOLUTION: There is provided a method for producing a contrast agent for optical imaging, which comprises at least the following steps: (a) a reaction step of reacting albumin and a specific organic coloring material or a salt thereof to obtain an aqueous solution containing a conjugate of the specific organic coloring material and albumin (b) a pH adjustment step of adjusting the pH of the aqueous solution containing the conjugate obtained in the (a) to 7.2 or less (c) a separation step of adding an ester solvent to the aqueous solution having a pH of 7.2 or less obtained in (b), followed by stirring it to separate into a layer of an organic solvent containing an ester solvent and a layer of an aqueous solution and (d) an extraction step of extracting an aqueous solution containing a conjugate from the solution obtained in (c) which is separated into a layer of an organic solvent and a layer of an aqueous solution (which may be also called an aqueous layer).SELECTED DRAWING: NoneCOPYRIGHT: (C)2017,JPO&INPIT【課題】 本発明では、アルブミンに結合していない単独の有機色素の残存量が少ない、逆に言えば、アルブミンと有機色素の結合体の割合が多い光学イメージング用造影剤の製造方法を提供することを目的とする。【解決手段】 本実施形態に係る光学イメージング用造影剤の製造方法は少なくとも以下の工程を有する。(a)反応工程:水溶液中で、アルブミンと、特定の有機色素又はその塩とを反応させて、この特定の有機色素とアルブミンとの結合体を含む水溶液を得る。(b)pH調整工程:(a)で得た結合体を含む水溶液のpHを7.2以下にする。(c)分離工程:(b)で得たpHが7.2以下の水溶液に、エステル溶剤を添加して撹拌することで、エステル溶剤を含む有機溶媒の層と、水溶液の層とに分離させる。(d)抽出工程:(c)で得た、有機溶媒の層と水溶液の層(水層と言うこともできる)とに分離した溶液から、結合体を含む水溶液を抽出する。【選択図】 なし
