The present invention relates to a macrocyclic compound, which specifically binds to a polo-box domain of polo-like phosphorylase-1, which inhibits mammalian oocyte maturation and represented by chemical formula 1, and to a composition for contraception containing the same, wherein the macrocyclic compound, according to the present invention, can be used for contraception by inhibiting oocyte maturation and resumption of meiosis after fertilization, and is safe from proteolytic enzymes as the compound is not in the form of a peptide analogue and has a significantly higher permeability of a zona pellucida and a biological membrane compared to drugs in the form of the peptide analogue.본 발명은 포유동물 난자 성숙을 저해하는 폴로 유사인산화효소-1의 폴로박스 도메인에 특이적으로 결합하는 화학식 1로 표시되는 거대 고리 화합물 및 이를 포함하는 피임용 조성물에 관한 것으로, 본 발명에 따른 거대 고리 화합물은 난자의 성숙 및 수정후 감수분열 재개를 억제함으로써 피임을 위한 용도로 사용할 수 있고, 펩티드 유사체 형태가 아니므로 단백질분해효소로부터 안전하고, 펩티드 유사체 형태의 약물에 비해 투명대와 생체막 투과성이 현저히 높은 효과가 있다.
