PROBLEM TO BE SOLVED: To provide a method for reducing the reactivity of ADAMTS-13 with an anti-ADAMTS-13 neutralizing antibody, and an ADAMTS-13 mutant which is produced by the method.SOLUTION: The method is characterized by substituting one of the following amino acid residues in charged amino acid residues located in a disintegrin-like domain, a cysteine-rich domain or a spacer domain of ADAMTS-13 by alanine: an arginine residue at position-312, a lysine residue at position-318, an arginine residue at position-568, a glutamic acid residue at position-569, an asparagine residue at position-579, an arginine residue at position-589, a lysine residue at position-608, an asparagine residue at position-614, a glutamic acid residue at position-634, an aspartic acid residue at position-635, an arginine residue at position-639, a glutamic acid residue at position-642, an arginine residue at position-644, a glutamic acid residue at position-651, an aspartic acid residue at position-654, an arginine residue at position-659, a glutamic acid residue at position-663, a glutamic acid residue at position-664, an asparagine residue at position-667, an arginine residue at position-670 and a lysine residue at position-681.【課題】ADAMTS-13と抗ADAMTS-13中和抗体との反応性を低減させる方法、並びに当該方法により作製されたADAMTS-13の改変体を提供する。【解決手段】本発明の方法は、ADAMTS-13のディスインテグリン様ドメイン、システインリッチドメイン又はスペーサードメインに存在する荷電アミノ酸のうち、下記アミノ酸の一つを、アラニンに置換することを特徴とする：第312位のアルギニン、第318位のリジン、第568位のアルギニン、第569位のグルタミン酸、第579位のアスパラギン、第589位のアルギニン、第608位のリジン、第614位のアスパラギン、第634位のグルタミン酸、第635位のアスパラギン酸、第639位のアルギニン、第642位のグルタミン酸、第644位のアルギニン、第651位のグルタミン酸、第654位のアスパラギン酸、第659位のアルギニン、第663位のグルタミン酸、第664位のグルタミン酸、第667位のアスパラギン、第670位のアルギニン、第681位のリジン。【選択図】なし
