The present invention provides antagonistic and agonistic anti-human VISTA antibodies as well as antibody fragments. These antagonist antibodies and antibody fragments can be used to inhibit or block the inhibitory effect of VISTA on T cell immunity, thereby promoting T cell immunity. These agonist antibodies and antibody fragments can be used to enhance or mimic the inhibitory effect of VISTA on T cell immunity, thereby suppressing T cell immunity. These antagonist antibodies and antibody fragments are particularly useful in the treatment of cancer and infectious conditions. These agonist antibodies and antibody fragments are particularly useful in the treatment of autoimmunity, allergies, inflammatory conditions, GVHD, sepsis, and transplant recipients.
