PROBLEM TO BE SOLVED: To improve the bioavailability of bosentan which is N-[6-(2-hydroxyethoxy)-5-(2-methoxyphenoxy)-2-(2-pyrimidinyl)-pyrimidin-4-yl]-4-tert-butylbenzene sulfonamide, is an endothelin receptor antagonist, and is used as a treating agent for pulmonary arterial hypertension by improving the solubility in water since bosentan is a sparingly soluble drug, and to obtain a method for industrially simply improving the bioavailability.SOLUTION: By making bosentan into amorphous bosentan by a solvent method, a melting method, a mechanochemical method or the like, the amorphous bosentan can be more simply produced in a shorter time than by the conventional production methods and furthermore, the solubility can be improved.COPYRIGHT: (C)2012,JPO&INPIT【課題】ボセンタンは、N-[6-(2-ヒドロキシエトキシ)-5-(2-メトキシフェノキシ)-2-(2-ピリミジニル)-ピリミジン-4-イル]-4-tert-ブチルベンゼンスルホンアミドであり、エンドセリン受容体拮抗薬を有し、肺動脈性高血圧症の治療剤として用いられている。ボセンタンは難溶性の薬物のため、水への溶解性を向上させることによるバイオアベイラビリティーの向上及びその工業的に簡便な向上方法が求められている。【解決手段】ボセンタンを溶媒法、溶融法、メカノケミカル法などによって非晶質ボセンタンとすることによって、従来の製造方法よりも簡便で短時間に製造でき、さらには溶解性を向上させることができる。【選択図】図4
