A method of determining an analyte concentration in a physiological sample, the method comprising: (a) introducing the physiological sample into an electrochemical cell comprising: (i) the first and second electrodes in a separate relationship； and (ii) a reagent layer comprising an enzyme and a mediator, (b) applying a first electrical potential that has a first polarity to the reaction cell and measuring the cell current as a function of time to obtain a first transient of time-current； (c) apply a second electrical potential that has a second polarity to the cell, in which the second polarity is opposite to the first polarity, and measure the cell current as a function of time to obtain a second time-current transient ； and (d) calculate a first current value based on an integral of current values with respect to a first time interval of the second time-current transient； (e) calculate a second current value based on an integral of current values with respect to a second time interval of the second time-current transient and a third current value based on an integral of current values with respect to a third time interval of the first time-current transient, in which the first, second and third time intervals are determined empirically so that the determined analyte concentration is accurate within ± 10% of a reference measurement, and has 2% variation in 1σ； (f) calculate a relationship comprising the second current value divided by the third current value； and (g) multiply the first current value minus a background value by the ratio to derive the analyte concentration.Un método de determinación de una concentración de analito en una muestra fisiológica, comprendiendo el método: (a) introducir la muestra fisiológica dentro de una celda electroquímica que comprende: (i) el primer y el segundo electrodos en una relación separada； y (ii) una capa de reactivo que comprende una enzima y un mediador, (b) aplicar un primer potencial eléctrico que tiene una primera pol
