The present disclosure relates to immunoregulatory fusion proteins containing an extracellular binding domain and an intracellular signaling domain, and target binding can generate a regulatory signal in a host cell such as a T cell. The present disclosure also relates to the use of immune cells expressing such immunoregulatory fusion proteins to treat certain diseases such as cancer or infectious diseases. In yet another aspect, the present disclosure is a method for treating a tumor, comprising administering to a subject having a tumor a therapeutically effective amount of a host cell described herein.
