Disclosed are methods of treating cancer with an MLKL modulator in vitro and in vivo. In some aspects, methods of treating small cell lung carcinoma, colon cancer, CNS cancer, melanoma, ovarian cancer, renal cancer, prostate cancer, and breast cancer are provided.
