Disclosed herein are methods of generating induced pluripotent stem cells. The method comprises providing a somatic or non-embryonic cell population, up-regulating the somatic or non-embryonic cell population with an amount of at least one reprogramming factor, an agent that down-regulates SIRT2, and / or up-regulates SIRT1. Contacting with the agent and culturing somatic or non-embryonic cells for a period of time sufficient to generate at least one induced pluripotent stem cell. Also provided herein are methods for differentiating cells by up-regulating SIRT2 and / or down-regulating SIRT1.
