The present specification relates to a culturing method of cultured root of Leguminous plants containing a high content of coumestrol, which can mass-produce coumestrol that exists in a minute amount in Leguminous plants. The culturing method comprises the following steps: (a) Leguminous seeds are germinated in a culture medium and induced into in-vitro plants having cotyledons, hypocotyls, and radicles； (b) one or more parts selected from cotyledons, hypocotyls, and radicles of the induced in-vitro plants are cultured in a culture medium and induced into a cultured root of each part； and (c) the induced cultured root of each part is cultured in a culture medium and propagated by adjusting the amount of air inflow and air composition, wherein the culture medium contains NH-4NO_3, CaCl_22H_2O, MgSO_47H_2O, KH_2PO_4, and KNO_3.본 명세서에는 콩과 식물에 극미량 존재하는 쿠메스트롤을 대량으로 생산할 수 있는 쿠메스트롤 함량이 증가된 콩과식물 배양근의 배양 방법에 관한 것으로, 상기 배양 방법은 (a) 콩과 식물 종자를 배양 배지에서 발아시켜 자엽, 배축 및 유근이 생성된 기내 식물체를 유도하는 단계； (b) 상기 유도된 기내 식물체의 자엽, 배축 및 유근 중 어느 하나 이상의 부위를 배양 배지에서 배양하여 부위별 배양근을 유도하는 단계； 및 (c) 상기 유도한 부위별 배양근을 배양배지에서 배양하며, 공기공급량 및 공기 조성을 조절하여 증식하는 단계； 를 포함하며, 상기 배양 배지는 NH4NO3, CaCl2 · 2H2O, MgSO4 · 7H2O, KH2PO4및 KNO3 의 영양성분을 포함하는 배지인, 방법에 관한 것이다.
