In the present description, a method is disclosed that describes a new technology that provides unsurpassed efficiency, flexibility, usefulness and speed of stable transgene incorporation into lymphocytes and other mammalian cells. The new method is based on the use of an mRNA-encoded transposase (for example, Sleeping Beauty transposase) in combination with a mini-ring encoded DNA moving element. The new method makes possible higher gene transfer rates and at the same time less toxic than the standard approach, which consists in using the encoded plasmid DNA transposase in combination with the encoded plasmid DNA moving element. Applications of the invention include, but are not limited to, the stable incorporation of a transgene encoding an immune receptor (eg, a T-cell receptor or a synthetic chimeric antigen receptor) into human T-lymphocytes, where the immune receptor provides specificity for the molecule expressed by the tumor cell.
