The present invention relates to a pharmaceutical composition and a cosmetic composition comprising an expression inhibitor of a RSK2 (ribosomal S6 kinase 2) gene or an activation inhibitor of a RSK2 protein as an active ingredient for preventing or treating skin cancer, and to a method for screening a medicine for skin cancer. According to the present invention, proliferation of skin cancer cells is inhibited through inhibition of RSK2 so that RSK2 can be used for targeted therapy of skin cancer.
