PROBLEM TO BE SOLVED: To provide TDP-43 variants having agglomerate-forming ability and cytotoxicity in the full length sequence, and methods for screening compounds for treating and/or preventing diseases associated with TDP-43 agglomerate accumulation using the TDP-43 variants.SOLUTION: The invention provides a TDP-43 variant comprising an amino acid sequence where cysteine at position 173 and/or position 175 in the amino acid sequence of TDP-43 is/are substituted. The invention also provides a method for screening a compound for treating and/or preventing a disease associated with TDP-43 agglomerate accumulation, which method comprises the steps of: (1) contacting the TDP-43 variant with a candidate compound； (2) detecting an activity of the compound to inhibit agglomerate formation； and (3) selecting a compound that inhibits the agglomerate formation.SELECTED DRAWING: Figure 6【課題】全長配列において凝集体形成能及び細胞毒性を有するTDP-43変異体、並びに該TDP-43変異体を利用したTDP-43の凝集体が蓄積する疾患を治療及び／又は予防するための化合物のスクリーニング方法を提供する。【解決手段】TDP-43のアミノ酸配列において、173位及び／又は175位のシステインが置換されたアミノ酸配列からなるTDP-43変異体、並びに以下の工程を含む、TDP-43の凝集体が蓄積する疾患を治療及び／又は予防するための化合物のスクリーニング方法：（１）該TDP-43変異体に候補となる化合物を接触させる工程、（２）該化合物の凝集体形成阻害活性を検出する工程、及び（３）凝集体形成を阻害する化合物を選択する工程。【選択図】図６
