The present invention relates to a composition for preventing and treating acetaminophen (AP)-induced hepatotoxicity, containing TNP(N2-(m-trifluorobenzyl) and N6-(p-nitrobenzyl)purine) as active ingredients. It is confirmed that TNP, which is known as an inhibitor for 5-inositol pyrophosphate, inhibits acetaminophen-induced cell death in human embryo stem cell-derived hepatocytes, mouse-derived hepatocytes and the human hepatoma cell line, increases reduced glutathione (GSH) concentration in hepatocytes, and inhibits phosphorylated JNK, which is a stress response increasing by acetaminophen, and it is also confirmed that the composition has the same protective effect against acetaminophen-induced hepatotoxicity as in the animal model, and thus TNP can be used as an active ingredient in the composition for preventing and treating acetaminophen-induced hepatotoxicity.La présente invention concerne une composition pour la prévention et le traitement de lhépatotoxicité induite par acétaminophène (AP), contenant du TNP (N2-(m-trifluorobenzyl) et N6-(p-nitrobenzyl) purine) en tant quingrédients actifs. Il est confirmé que le TNP, qui est connu en tant quinhibiteur de pyrophosphate 5-inositol, inhibe la mort cellulaire induite par acétaminophène dans des hépatocytes dérivés de cellules souches dembryon humain, dhépatocytes dérivés de la souris et la lignée cellulaire dhépatome humain, augmente la concentration de glutathione réduite (GSH) dans les hépatocytes, et inhibe le JNK phosphorylé, qui est une réponse au stress augmenté par lacétaminophène.
