Disclosed are imidazo[1,2-a]pyridine derivatives as represented by the general formula (I), wherein: T and U are independently chosen from CR3 and CRA, such that exactly one of T and U is CRA and V is N Z1, Z2 and Z3 are each CH W is -C(=O)NR4-, -NR4C(=O)- or -NR4-NR4-C(=O)- X is absent or is an optionally substituted alkylene group Y is cycloalkyl, heterocycloalkyl, aryl or heteroaryl, each of which may be substituted and wherein the remaining substituents are as defined herein. Further disclosed is a pharmaceutical composition, which comprises at least one compound as defined above, or a salt or hydrate thereof in combination with a physiologically acceptable carrier or excipient for modulating the activity of a P2X7 receptor, and particularly for the treatment of pain, inflammation, a neurological or neurodegenerative disorder, a centrally-mediated neuropsychiatric disorder, a cardiovascular disorder or an immune system disorder, osteoarthritis, rheumatoid arthritis, lupus erythematosus, multiple sclerosis, arthrosclerosis, glaucoma, irritable bowel syndrome, inflammatory bowel disease, Alzheimers disease, traumatic brain injury, asthma, chronic obstructive pulmonary disease, or interstitial fibrosis.
