PURPOSE: A human monoclonal antibody-derived bispecific antibody and a composition containing the same are provided to simultaneously neutralize VEGFR-2 and c-Met and to effectively suppress signal transduction mechanism of angiogenesis and cancer cell proliferation.CONSTITUTION: A bispecific antibody with binding specificity to VEGFR-2(vasicular endothelial growth factor receptor-2) and c-Met(mesenchymal-epithelial transition factor) contains water soluble peptides of sequence numbers 21 or 22, which is linked with the N-terminal of a light chain of an endothelial growth factor receptor neutralizing antibody. The antibody contains a light chain variable region of sequence numbers 1-19 and a heavy chain variable region of sequence number 20. The antibody is scFv(single chain fragment variable) or IgG(immunoglobulin g). The linker contains a base sequence of sequence number 23.COPYRIGHT KIPO 2013본 발명은 혈관신생에 관여하는 VEGFR-2 및 c-Met을 동시에 중화시킴으로써, 혈관신생 및 암세포 증식에 관련된 신호전달기작을 효과적으로 억제할 수 있는 인간 단일클론항체 유래 이중표적항체와 상기 항체를 함유하는 혈관신생 억제 및 암 치료용 조성물을 제공한다. 본 발명에 따른 이중표적항체는 혈관신생과 암세포 증식에 관여하는 두 가지 표적을 동시에 중화시킴으로써, 기존의 단일표적 항체에 비해서 뛰어난 중화능을 보일 뿐만 아니라, 암 치료에도 매우 효과적이다. 또한 상호 연관성이 있는 두 가지 표적에 대하여, 본 발명자가 발명한 형태의 이중표적항체를 제작함으로써 단일표적 항체 처치에 의한 실익보다 우수한 효과를 기대할 수 있다.
