Method for transforming Dendranthema X grandiflorum using petal explant of Dendranthema X grandiflorum, and transgenic Dendranthema X grandiflorum prepared thereby
PURPOSE: A transgenic Dendranthema X grandiflorum prepared by a petal explant of Dendranthema X grandiflorum is provided to enhance transformation efficiency. CONSTITUTION: A method for transforming Dendranthema X grandiflorum using a petal explant of Dendranthema X grandiflorum comprises: a step of washing and disinfecting Dendranthema X grandiflorum; a step of separating and cutting petals from Dendranthema X grandiflorum; a step of culturing the cut petals in a medium(SIM) for two days; a step of shaking Agrobacterium containing pCAMBIA 2301 with GUS gene in YEP medium; a step of dipping the petal explant in the Agrobacterium suspension(O.D. 0.8) for 20-50 minutes; a step of moving the inoculated petal explant to SIM and co-culturing for two days; a step of washing the petal explant in MS liquid medium and culturing in a primary selection medium; and a step of selecting redifferentiated Dendranthema X grandiflorum four weeks later and culturing in a rooting inducing medium(RIM).
