The present invention relates to a drug delivery field, and particularly, to a cationic liposome-based drug delivery field. In an embodiment, the present invention provides a method for preparing ligand-targeted (for instance, antibody- or antibody fragment-targeted) liposomes which are useful to deliver liposomes to a tumor including brain tumor. In an embodiment, the liposomes are delivered by delivering temozolomide across a blood-brain barrier to treat primary or metastatic brain tumor. Additional cancer which can be treated with liposomes includes neuroendocrine tumor, melanoma, prostate cancer, head and neck cancer, ovarian cancer, lung cancer, liver cancer, breast cancer, urinogenital cancer, stomach cancer, colorectal cancer, cervical cancer, liposarcoma, rhabdomyosarcoma, choriocarcinoma, pancreatic cancer, retinoblastoma and other types of cancer. In another embodiment, the liposomes deliver melphalan to treat multiple myeloma, other blood tumors, or other solid tumors.
