PROBLEM TO BE SOLVED: To provide BMP-ALK3 antagonists and uses thereof for promoting bone growth.SOLUTION: In certain aspects, the invention provides a composition and method for promoting bone growth and increasing bone density and strength. In certain embodiments, the invention provides ALK3 polypeptides including ALK3-Fc fusion proteins. The invention demonstrates that a soluble form ALK3 acts as an inhibitor of BMP-ALK3 signaling, and promotes increase in bone density, bone growth and bone strength in vivo.
