A transgenic mouse having a genome comprising a homozygous disruption of a common receptor γ gene (Il2rg -1-), a transgene encoding signal regulatory protein a (SIRPα) that is functionally expressed in the mouse, and wherein the mouse genome further comprises a transgene encoding thymic stromal cell-derived lymphopoietin (TSLP) that is functionally expressed in the mouse or wherein exogenous thymic stromal cell-derived lymphopoietin (TSLP) is administered to the transgenic mouse. A Human Immune System (HIS) mouse made from said transgenic mouse. Use of the HIS mouse as a preclinical mouse model for testing human vaccine candidates, immunotherapeutics or anti-infectious agents. The HIS mouse as a spontaneous model of human atopic dermatitis for testing novel therapeutics.Une souris transgénique ayant un génome comprenant une interruption homozygote d'un gène γ de récepteur commun (Il2rg -1-), un transgène codant pour une protéine régulatrice de signal a (SIRPα) qui est exprimé de manière fonctionnelle chez la souris, et le génome de souris comprenant en outre un transgène codant pour la lymphopoïétine dérivée de cellules stromales thymiques (TSLP) qui est exprimé de manière fonctionnelle chez la souris ou une lymphopoïétine dérivée de cellules stromales thymiques exogènes (TSLP) étant administrée à la souris transgénique. L'invention concerne également une souris de système immunitaire humain (HIS) produite à partir de ladite souris transgénique. L'utilisation de la souris HIS en tant que modèle de souris préclinique pour tester des candidats de vaccins humains, des agents immunothérapeutiques ou des agents anti-infectieux. La souris HIS est un modèle spontané de dermatite atopique humaine pour tester de nouveaux agents thérapeutiques.
