The present invention relates to a method for screening an anticancer agent for inhibiting a bond of methionyl-tRNA synthetase (MRS) and cyclin-dependent kinase 4 (CDK4), and to an anticancer composition including an anticancer agent sorted through the method as an active ingredient. More specifically, the method comprises the steps of: making MRS or a fragment thereof come in contact with CDK4 or a fragment thereof while a test material exists or does not exist measuring a bond of MRS and CDK4 determining a change in a bonding level of MRS and CDK4 by a test material sorting a test material for reducing a bonding level of MRS and CDK4 and confirming an anticancer activity of the sorted test material in a cell or an animal. The present invention can be useful in developing an anticancer agent having a totally novel operation mechanism on the basis of a discovery that MRS is a novel regulator of CDK4 for directly binding with and stabilizing CDK4 which is important for the occurrence and the progress of cancer.
