The present invention relates to a sorting vector for sorting cells, wherein the sorting vector includes exogenous target genes and to a method for sorting cells by using the same. The sorting vector (pEF1/HisC:VEGF:EGFP) of the present invention includes VEGF and EGFP genes, and the expression of each of the genes is independently adjusted by means of an EF promoter and a CMV promoter. VEGF-transduced MSCs sorted according to the present invention has the increased expression of a vascular endothelial growth factor, and angiogenesis is induced. Thus, an ischemic lower body part treating effect is improved. A cell sorting system using a sorting vector of the present invention has following advantages compared to typical cell-based gene delivery. Gene delivery efficiency is high even at a low concentration. The cell sorting system can be useful in treating severe ischemic diseases requiring a large quantity of angiogenesis factors in a short time. Target genes or base cells can be easily replaced, so the system can be applied to various diseases using cell-based gene treatment.COPYRIGHT KIPO 2017본 발명은 외인성(exogenous) 대상 유전자를 포함하는 세포를 선별하기 위한 선별 벡터 및 이를 이용한 세포 선별 방법에 관한 것이다. 본 발명의 선별벡터(pEF1/HisC:VEGF:EGFP)는 VEGF 및 EGFP 유전자를 포함하며, 각각 유전자의 발현은 EF 프로모터 및 CMV 프로모터에 의해 독립적으로 조절된다. 본 발명에서 선별된 VEGF-형질도입된 MSCs 는 혈관내피성장인자의 발현이 증가되어 신생혈관의 생성을 유도함으로써 허혈성 하지 치료효과를 향상시킨다. 본 발명의 선별벡터를 이용한 세포선별 시스템은 전형적인 세포-기반 유전자 전달과 비교하여 다음과 같은 이점이 있다: 저농도의 세포에서도 유전자 전달 효율이 높다 단기간에 많은 양의 혈관신생 인자가 요구되는 중증 허혈성 질환의 치료에 유용하게 사용될 수 있다 타겟 유전자 또는 기반세포의 교체가 용이하여 세포-기반 유전자치료를 이용하는 다양한 질병에 적용될 수 있다.
