Předmětem předloženého řešení je způsob přípravy apixabanu vzorce I, při kterém reaguje ethyl 6-(4-jodfenyl)-1-(4-methoxyfenyl)-7-oxo-4,5,6,7-tetrahydro-1H-pyrazolo[3,4-c]pyridin-3-karboxylát vzorce III s piperidin-2-onem vzorce IV v přítomnosti báze a ligandu a za katalýzy mědí nebo mědnatými ionty, kde se jako báze použije sůl kyseliny fosforečné a jako ligand se použije amin ze skupiny 1,2-diaminů v aprotickém rozpouštědle, zvoleném ze skupiny etherů obecného vzorce R.sup.1.n.-O-R.sup.2.n., výhodně cyklopentylmethylether, a připraví se ethyl 1-(4-methoxyfenyl)-7-oxo-6-[4-(2-oxo-1-piperidin-1-yl)fenyl]-4,5,6,7-tetrahydro-1H-pyrazolo-[3,4-c]pyridin-3-karboxylát, který se převede reakcí s amoniakem ve vhodném rozpouštědle na apixaban vzorce I, který se izoluje a případně krystaluje.The present invention relates to a process for preparing apixaban of the general formula I, comprising reacting ethyl 6-(4-iodophenyl)-1-(4-methoxyphenyl)-7-oxo-4,5,6,7-tetrahydro-1H-pyrazolo[3,4-c]pyridine-3-carboxylate of the general formula III with piperidin-2-one of the general formula IV in the presence of a base and a ligand and under catalysis of copper or copper ions, wherein the base used is a phosphoric acid salt and the ligand used is an amine being selected from the group consisting of 1,2-diamines in aprotic solvent, being selected from the group consisting of ethers of the general formula Re1-O-Re2, preferably cyclopentyl methyl ether, wherein there is prepared ethyl 1-(4-methoxyphenyl)-7-oxo-6-[4-(2-oxo-1-piperidin-1-yl)phenyl]-4,5,6,7-tetrahydro-1H-pyrazolo-[3,4-c]pyridine-3-carboxylate, which is then converted by the reaction with ammonia in a suitable solvent to apixaban of the general formula I, which is then isolated and optionally subjected to crystallization.
