PURPOSE: A method for preparing silk fibroin of high molecular weight is provided to reduce blood glucose and cholesterol. CONSTITUTION: A composition for reducing blood glucose or cholersterol contains silk fibroin of high molecular weight as an active ingredient. The silk fibroin is 10%(w/w) silk formic acid solution with 70-350 of viscosity. The composition further contains sericin as an active ingredient. A health food for relieving or preventing diabetes, obesity, or hyperlipidemia contains the composition. A method for preparing the silk protein comprises: a step of refining silk cocoon and refining in distilled water at 90-100°；C for 1-3 hours； a step of washing the refined silk cocoon using distilled water at 60-80°；C； a step of dissolving the washed silk cocoon with a mixture solvent of LiBr solution or CaCl_2/water/ethanol； a step of removing salt by a cut-off tube； and a step of freeze-drying the silk protein solution.본 발명은 혈당 및 콜레스테롤 강하특성이 있는 고분자량의 실크 피브로인 및 그 제조방법에 관한 것으로, 더욱 구체적으로 19만-40만의 평균분자량(Da)을 갖는 고분자 실크 피브로인 및 임의적으로 세리신을 유효성분으로 포함하는 혈당 또는 콜레스테롤 강하용 조성물 및 그 제조방법에 관한 것이다.본 발명에 따르면, 저분화된 실크 피브로인 펩타이드가 아닌 고분자량의 피브로인이 오히려 더 뛰어난 혈당 및 콜레스테롤 저하 효과를 나타내고, 이러한 고분자량의 피브로인에 세리신을 조합함으로써 더욱 우수한 혈당 및 콜레스테롤 저하 효과를 나타내므로, 당뇨병, 비만, 또는 고지혈증 치료 또는 예방용 기능성 식품 또는 의약품의 제조에 효과적으로 이용될 수 있다. 또한, 기존 실크 단백질의 제조공정에서 세리신을 제거하는 공정이나 피브로인을 저분화하는 공정을 생략할 수 있어서 전체적인 제조원가를 절감할 수 있는 장점이 있다.
