The present invention discloses a dual-chamber mixing device for a syringe and its assembly method, and a syringe using the same and its manufacturing and operation method. The dual-chamber mixing device comprises an outer barrel and an inner barrel concentrically arranged to form an outer chamber, the inner barrel having an inner chamber. A mixing plunger is slidably arranged in the outer chamber. A seal located in the outer chamber is axially movable, in response to depression of the mixing plunger, from a first position in sealing engagement with one or more apertures in the inner barrel to a second position between the apertures and vents in the outer barrel, thereby allowing the depression of the mixing plunger to force a first mixed substance from the outer chamber through the apertures to be mixed with a second mixed substance in the inner chamber.
