This invention relates to a manufacturing method of substrate having anti-fog coating layer and the constituent structure thereof. The method uses cellulose acetate to form an anti-fog coating layer of a predetermined configuration and thickness on the substrate of lens, wherein the thickness is better lying between 0.1-0.3 mm in practice, and performs lamination of the anti-fog coating layer with the electroplating-treated lens (i.e. the substrate layer) to constitute the substrate of lens.
