The present disclosure relates to combinations of RET inhibitors with mTORC1 inhibitors and their use in the treatment of various cancers mediated by aberrant RET activity. Preferably, the cancer is selected from thyroid cancer, non-small cell lung cancer, paraganglioma or intrahepatic cholangiocarcinoma. The RET inhibitor is preferably BLU-667 and the mTORC1 inhibitor is preferably everolimus or AZD8055.
