Mapracorat dose dependently inhibited the TNFa secretion from activated canine PBMC with an IC50 value of approx. 0.2 nmo1/1. Intradermal injection of compound 48/80 (50 μg in 50 μ1 saline), resulted in a clear wheal and erythema reaction over the 60 min observation period in vehicle (acetone) treated areas. Topical pre-treatment with mapracorat (0.1%) and triamcinolone acetonide (0.015%) led to significant reduction in wheal and erythema area. A once daily topical administration of triamcinolone acetonide significantly reduced skin fold thickness from day 8 to day 14, whereas no such reduction was observed for mapracorat. Taken together, in vitro and in vivo experiments demonstrate that mapracorat has comparable antiinflammatory efficacy to triamcinolone acetonide, while skin fold thickness indicates a better safety profile compared to steroidal glucocorticoids at equi-effective dosages.
