PROBLEM TO BE SOLVED: To provide a peptidomimetic macrocycle using p53 which regulate the activity of p53 by inhibiting the interaction between p53 and HDM2, p53 and HDMX, or p53 and proteins of both HDM2 and HDMX, and which can be used for treating diseases including (but not limited to) cancer and other hyperproliferative diseases.SOLUTION: The p53 peptidomimetic macrocycle is a peptide comprising a part of stably crosslinked human p53, the crosslinked peptide contains at least two modified amino acids, the intramolecular crosslinking being formed to stabilize the α-helix secondary structure of a part of p53 thought to be important for binding p53 to HDM2 and p53 to HDMX.SELECTED DRAWING: None【課題】ｐ５３及びＨＤＭ２、ｐ５３及びＨＤＭＸ、又はｐ５３及びＨＤＭ２とＨＤＭＸ両方のタンパク質との間の相互作用を阻害することでｐ５３の活性を調節し、かつ、癌及び他の過剰増殖性疾患（ただし、これらには限定されない）を含む疾患の治療に使用することが可能な、ｐ５３を利用したペプチド模倣大環状分子の提供。【解決手段】安定に架橋されているヒトのｐ５３の一部からなるペプチドであり、架橋ペプチドは修飾されたアミノ酸を少なくとも２つ含み、ｐ５３のＨＤＭ２への結合及びｐ５３のＨＤＭＸへの結合に重要だと考えられているｐ５３の一部のα－ヘリックス二次構造を安定化できる分子内架橋を形成した、ｐ５３ペプチド模倣大環状分子。【選択図】なし
