Haptens, immunogen and tracers of structure I wherein R is either benzyl or isobutyl； two of X1, X2, and X3 are H and the other is for the hapten SH-X- in which X is a straight chain or substituted straight chain or branched alkyl group, preferably of 1-10 carbon atoms, for the immunogen accm-spacer- where accm is an antigenicity conferring carrier material, for the tracer HRP-spacer where HRP is horseradish peroxidase. Preferably, X1 = X3 = H and X2 is HS-CH2-CH(NH2)-C(O)-. In another aspect, an immunoassay method which detects the antibiotics Colistin and Polymyxin B. A targeted synthetic method produces novel haptens and immunogens enabling the production antibodies sensitive to each antibiotic. Also described are kits and tracers.
