A method of reducing the production of lactate in cultured cells, the method comprising culturing cells containing a first heterologous nucleic acid sequence encoding a small interfering RNA (siRNA) specific for lactate dehydrogenase (LDH) and a second heterologous nucleic acid sequence encoding a siRNA specific for pyruvate dehydrogenase kinase (PDHK), where the sequence of the first heterologous nucleic acid is functionally linked to the first promoter and the second gete sequence ologichnoy nucleic acid is operably linked to a second promotorom.2. The method of claim 1, wherein the LDH is LDHa. The method of claim 1, wherein the cultured cells further comprise a third heterologous nucleic acid sequence encoding siRNA specific for the second PDHK, and the third heterologous nucleic acid sequence is operably linked to the third promoter. The method of claim 3, wherein the cultured cells further comprise a fourth heterologous nucleic acid sequence encoding siRNA specific for the third PDHK, and the fourth heterologous nucleic acid sequence is operably linked to the fourth promoter. The method of claim 4, wherein the cultured cells further comprise a fifth heterologous nucleic acid sequence encoding siRNA specific for the fifth PDHK, and the fifth heterologous nucleic acid sequence is operably linked to the fifth promoter. A method according to any one of claims 1, 3, 4 and 5, wherein the PDHK is selected from the group consisting of PDHK1, PDHK2, PDHK3 and PDHK1.
